Since I'm using a Bambu Lab printer, I'm not sure if the process is analogous. My printer prints a color on layer one, then switches to a new color on layer one, and so forth, up to five colors, then moves to layer two. Other than loading the filaments I need, I don't do anything. The print profile either supports AMS or not. Some models have a separate profile for each option, in case you don't have the AMS, though probably not signs or other flat things.

Radio Shack used to carry whole sheets of rub-on letters / numbers. You had to line up each letter / number manually. You'd put the sheet over the surface and use a pencil to run the material onto the surface. Of course, if you didn't spray a thin coat of acrylic clear-coat, the lettering would eventually come off. Here's an example.
